Résumé
A major obstacle in the development of effective cellular therapies for solid malignancies has been the identification of differentially expressed cell surface proteins that are required for tumor susteneance. Her ewe will discuss recent advancs in immunopeptidomics that has resulted in the identification of non-mutated peptides derived from epigenetically deregulated intracellular oncoproteins recurrently presented on common HLA alleles that can be targeted with synthetic immunotherapies. Due to the peptide-centric nature of the binders, HLA restriction is relaxed comparted to TCRs, and potent killing has been observed across HLA allotypes in preclinical models. Plans for an initial clinical trial in the childhood cancer neuroblastoma and ongoing discovery and development efforts in other cancers will be discussed, including the importance of CAR-pMHC complex structure in derisking cross reactivity liabilities.
