Multiomic approaches to understand mechanisms of Down syndrome pathogenesis to develop therapeutic interventions

Résumé
The chromosome 21, in trisomy in Down syndrome (DS) individuals, harbors several immunological genes, including the ones triggered by SARS-CoV-2 infection. We and others have shown that part of the SARS-CoV-2 triggered gene signatures are elevated at baseline in uninfected DS individuals. The application of Multi-Omic platforms can support the mechanistic understanding of major pathways and upstream signals associated to this heightened inflammation in the trysomic individual, thus guiding the development of precise therapeutic interventions, possibly ameliorating or reversing inflammation related comorbidities. We hypothesize that the circulating microbiome and its metabolites, their subsequent role in epigenetic alterations, and further modulation of gene expression of innate and adaptive immune responses could be a potential upstream mechanism leading to high basal levels of inflammation in part of the DS subjects, resulting in different levels of comorbidities, and more severity post-SARS-CoV-2 infection.
